Recent evidence from our laboratory indicates that both epidermal growth factor (EGF) and fibroblast growth factor (FGF) stimulate the migration of human glioma cells through highly organized membrane protrusions that we have termed pseudopods. The activation of these protrusions is highly dependent upon actin polymerization and requires the concerted actions of small G-proteins, guanine nucleotide exchange factors and phospholipase C (PLC). The objective of this project is to further define the relationship between these molecules and the regulation of pseudopod formation and their role in the motility of human glioma cells. This project will further determine the cellular location and activation kinetics of the different signaling molecules and their relationship with the actin cytoskeleton. In Specific Aim 1, the hypothesis that phosphatidylinositol (PI) 3-kinase is required for pseudopod assembly and protrusion formation will be tested. Experiments in this aim will examine the possible interaction between PLC and PI 3-kinase in the regulation of pseudopod formation and cell motility. The effects of PLC inhibitors on the localization of PI 3-kinase and the actin cytoskeleton in glioma cells will be examined. A similar approach will be used to determine whether PLC is necessary for pseudopod formation in a glioma cell line overexpressing constitutively active ras. In Specific Aim 2, the hypothesis that members of the Ras family of G-proteins, specifically Ras, Rap1, and R-Ras2, are necessary for the assembly and protrusion of pseudopods will be tested.
